Abstract
Bakgrunn: Overvekt, fedme og diabetes type 2 (T2D) er et stadig økende problem i dagens verdenssamfunn. Akkumulering av triacylglyserol (TAG) kan være assosiert med fedme og T2D, og kan føre til ugunstige effekter på energimetabolismen. Diacylglyserol acyltransferase (DGAT) 1 og 2 katalyserer det siste trinnet i triglyseridsyntesen; dannelsen av TAG fra diacylglyserol (DAG). Fokuset i denne oppgaven er å studere om DGAT1 og DGAT2 har ulike roller i lipidmetabolismen i humane skjelettmuskelceller (myotuber) ved å bruke selektive hemmere av DGAT1 og DGAT2. Metode: Humane satelittceller ble isolert fra biopsier fra musculus vastus lateralis (den ytre brede lårmuskelen), dyrket og differensiert til myotuber. Disse ble behandlet med hemmere av DGAT1(A922500) og DGAT2(JNJ-DGAT2-A), med og uten eksogene substrater. Energimetabolismen ble undersøkt ved å benytte [1-14C]oljesyre, D-[14C(U)]glyserol og D-[14C(U)]glukose. Lipiddistribusjonen, ved hjelp av inkorporering av radiomerket oljesyre, glyserol og glukose i TAG og andre lipidklasser, ble bestemt ved tynnsjiktkromatografi. Fettsyreoksidasjonen ble studert ved kvantifisering av syreløselige metabolitter (ASM) og CO2. Resultater: Konsentrasjons-responskurve med DGAT1-hemmer viste at 1 μM ga en effektiv hemming av dannelsen av TAG. Ved bruk av DGAT1-hemmer ble inkorporeringen av 14C-oljesyre i ulike lipidklasser redusert, samt at oljesyreoksidasjonen økte. Ved bruk av DGAT2-hemmer økte syntesen av TAG og oljesyreoksidasjonen ble redusert. Inkorporering av 14C-glyserol i ulike lipidklasser ble redusert ved DGAT2-hemming i fravær av eksogene oljesyre, mens i nærvær av oljesyre ble inkorporeringen av 14C-glyserol i TAG kun redusert ved DGAT1-hemming. Lipogenesen fra 14C-glukose stimulert av LXR-aktivator ble redusert etter DGAT2-hemming. Konklusjon: Selv om DGAT1 og DGAT2 katalyserer samme reaksjon i triglyseridsyntesen, ser det ut til at de har ulike roller i skjelettmuskulatur. Resultatene i oppgaven antyder at DGAT1 er ansvarlig for inkorporering av eksogene fettsyrer til TAG, mens DGAT2 potensielt spiller en viktigere rolle ved lipogenese (fettsyresyntese) og inkorporering av endogene fettsyrer i TAG. Resultatene tyder også på at dannelsen av TAG har flere ”pools” og at det er flere metabolismeveier til fettsyreoksidasjon
Background: Overweight, obesity and type 2 diabetes (T2D) is an increasing problem in today´s world society. Accumulation of triacylglycerol (TAG) may be associated with obesity and T2D, and may adversely affect the energy metabolism. Diacylglycerol acyltransferase (DGAT) 1 and 2 catalyses the final step in the triglyceride synthesis; formation of TAG from diacylglycerol (DAG). In this thesis, we have studied whether DGAT1 and DGAT2 have different roles in lipid metabolism in human skeletal muscle cells (myotubes) using selective inhibitors for DGAT1 and DGAT2. Methods: Human satellite cells were isolated from biopsies of musculus vastus lateralis (the outer broad thigh muscle), cultured and differentiated to myotubes. The myotubes were treated with DGAT1 (A922500) and DGAT2 (JNJ-DGAT2-A) inhibitors, with or without exogenous substrates. Concentration response was studied using different concentrations of the DGAT1 inhibitor. Energy metabolism was then investigated using radiolabeled [1-14C]oleic acid, D-[14C (U)]glycerol and D-[14C (U)]glucose. Lipid distribution was determined by thin layer chromatography and fatty acid oxidation was measured as formation of acid-soluble metabolites (ASM) and CO2. Results: Concentration response curve with DGAT1 inhibitor showed that 1 μM gave an effective inhibition of TAG formation. DGAT1 inhibitor reduced the incorporation of 14C-oleic acid into different lipid classes, while fatty acid oxidation was increased. However, by inhibiting DGAT2 we observed increased TAG formation and reduced oxidation from 14C-oleic acid. Incorporation of 14C-glycerol into different lipid classes was reduced to a greater extent by DGAT2 inhibition compared with DGAT1 inhibition in the absence of exogenous oleic acid. With exogenous oleic acid added, the incorporation of 14C-glycerol by DGAT1 inhibition was reduced compared to DGAT2 inhibition. Lipogenesis from 14C-glucose stimulated by a LXR-activator was reduced after DGAT2-inhibition. Conclusion: Although DGAT1 and DGAT2 catalyse the same reaction in triglyceride synthesis, they may have different roles in skeletal muscle. The results from this thesis suggest that DGAT1 is responsible for incorporation of exogenous fatty acids into TAG, while DGAT2 potentially plays an important role in lipogenesis (fatty acid synthesis) and incorporation of endogenous fatty acids into TAG. Further results also indicate that the formation of TAG has more “pools”, and that there are multiple pathways to fatty acid oxidation.
